Since the software upgrade, the open cash drawer button is greyed out. Even with full access rights, the button remains greyed out.
Any clues as to why, and how do I fix?

When you say "even with full access rights", is that because you're logging on with a team passcode? Or are you logging on as the account owner?
Would you be able to let me know what device you're using to access the app?
While I await your response, there are a few basic troubleshooting steps I can get you to try to resolve minor bugs like this:
1. Ensure your device operating system is up-to-date.
2. Delete and reinstall the Point of Sale app on your device.
3. Double-check your Cash Management settings by navigating to ≡ More > Add-ons > Cash Management > Add to Point of Sale. If you have already added Cash Management, it should show in your settings.
I recommend reviewing our Cash Management guide to ensure you've enabled the correct settings in the app and for any team members as required.

After consulting with my team, it seems there's a known issue similar to this happening on both Square Registers and Square Terminals. A solution is currently being deployed to sellers in the upcoming app update (version 6.38), scheduled to be available by the end of the day tomorrow. (Tuesday, March 12th AEDT).
I suggest updating to the latest version of the app once it becomes available tomorrow and testing to ensure the cash management functionality is restored. If the issue persists beyond tomorrow, please reach out to our Support Team for further assistance, and they'll be happy to take a closer look at your device with you.
