If you set the customer up as taxable, then yes, they will be charged tax automatically at the sale (for items which you set up as taxable.) You can manage your sales taxes from the mobile Dashboard by tapping on the three horizontal lines, then Settings, then Taxes. Add a new tax by tapping on the '+' in the upper right corner or edit an existing tax by tapping on it.
