Under Taxes in your dashboard you will have to make sure the Tax rates are available on your items and location. Then when they are ringing up items they will have to apply the correct tax to each item.
The better way to do this would be to create a seperate location. This way location A is at one tax rate and Location B is at another. But creating a seperate location has other advantages and possible disadvantages.
