I created a digital product in item library and offered it as a subscription. I am trying to remove the shipping charge but unsuccessful

I would recommend checking the Item Preferences section of your Square Online Dashboard. Under Default item fulfillment, make sure you do not have shipping selected.
You can locate our Item Preferences by going to your Square Online Dashboard> Items>Item preferences>Default item fulfillment.

I did change the setting but that did not work, what I realized is that I am selling the Membership (Digital Item) on my POS system, I do not have an online sales channel on Squareup. Is there any way to remove the shipping?

Currently, subscriptions can only be fulfilled via shipping.
In order to create a subscription that allows you to sell items, you'll need to select shipping as a fulfillment method.
The fulfillment setting you have entered on the Square Online overview page will apply to all Payment Links.
