I want to sell some one off items. Some items there will be one of, others may have multiples available, but once they are gone, they are gone. At the moment my online store will continue to allow an item to be purchased even once the inventory is zero. How do I set it so once inventory is zero, the item can no longer be purchased?

Provided you're managing stock levels via the Square dashboard, and have synced your location inventory to Square Online this should occur automatically.
