Customers not receiving order ready emails

We are doing pick-ups five days a week for our site. To make this work, I'm banking on sending the automated "Your order is ready" emails. I thought this happened whenever I clicked on "Mark Order As Ready." However, none of my customers are receiving notifications when I click that button. Am I doing something wrong, or is something not working?
