I have been using readers for several years but today when I went to set up my phones I got the same message that "rooted devices are not supported". Two different phones and two readers but nothing working. Worked fine last week and developer options are not open on either phone. Any ideas ?

That definitely sounds strange if everything was working well last week.
Can you let me know the brand and model of both phones?
Can you try the following steps on both devices when you have a chance and let me know if the error message persists? Factory resetting the Readers may help.
Open Square Point of Sale App:
1) Settings > Hardware > Card Readers > forget all card readers
2. Force-quit Square Point of Sale App
3. Open device settings
4. Navigate to Bluetooth
5. Forget all Square Readers
6. Hold the button on the reader until the lights flash red
7. Reboot the Device
8. Re-launch Square Point of Sale app, wait 1-2 minutes, and re-pair the reader
