As soon as the charged/debited transaction is finalized, that amount (minus Square's processing fees) will be available on your Square card. Here's a thread where this question is answered by a Square representative. Note the final reply, where it's stated that once you activate your card, automatic deposits are suspended so if you want to transfer funds to your linked bank account, you'll need to do that manually.
