You can add a SKU to any item within your item library in your Square Dashboard or Point of Sale app. Here is a good article on Inventory Management by Square, which goes over item creation and adding SKUs, it states:

"From the Square Point of Sale app:

Tap the three horizontal lines > Items.
Create or select an existing item.
Enter a stock amount under the item details or tap Prices, Sizes, or SKUs to add stock to an item with multiple variations."

If performing this action from the Square Dashboard, use the item library link provided above. (Items > Items Library)

If you eventually want to create/add barcodes for your SKUs, here is a thread on barcodes you might find helpful.
